Abstract: Dermal papilla cells (DPCs) are the critical signaling center that orchestrates hair follicle (HF) neogenesis by the interaction with the microenvironment, such as hair matrix cells (HMCs), dermal sheath cup cells (DSCCs), and the extracellular matrix (ECM). An optimized three dimensional (3D) biomimetic co-culture system was developed to restore the inductive characteristics of DPCs and investigate the intercellular interaction with other cells. To determine why the 3D biomimetic co-culture system of DPCs afford better biological function, we investigated the mRNA expression profiles of DPCs-P4 (Control, P4-DPCs cultured in 2D environment as negative control), DPAs-3D model (Model), DPAs-Bio system (System), DPCs-P0 (P0) and using high-throughput sequencing. 12 Samples (3 samples for each group) were collected from three individuals.
