RNAseq anaysis of hippocamal parvalbumin-positive interneurons in mice exposed to chronic social defat stress

Abstract: Purpose: To identify genes differentially expressed in parvalbumin-positive (PV) interneurons of the hippocampus in non-defetaed (CTRL), stress-susceptible and stress-resilient mice. Method: Translating Ribosome Affinity Purification (TRAP) to isolate RNA from PV+ cells, cDNA synthesis and next generation RNAseq using Illumina Nextseq 500 sequencer. Results: RNA-seq revealed 458 DEGs between non-defeated and resilient mice, 1976 DEGs in non-defeated and susceptible mice, and 3475 DEGs in resilient and susceptible mice. Overall design: Cre-dependent EGFP-L10a mice crossed with PV-cre mice (EGFP-L10a+; PV-Cre+) were exposed to chronic social defeat stress. Two hippocampi from one mouse were pooled for one TRAP sample. CTRL is a TRAP/RNAseq dataset for non-defeat group. TRL Input is RNAseq dataset for the total RNA sample of non-defeat group before TRAP (translating ribosome affinity purification) procedure.
